The developers have actually stated that it will be the most persistent MMO in existence. There won't be any quests in the game, rather Events that anyone can partake in. That's what I call immersion.
Instances will be used for important storyline shifts that could not take place in a persistent world. The limitations of doing missions in a persistent world would suck and make them like any old quest in other MMO's. Have the very large majority of the world persistent, whilst reserving instances for missions and dungeons only is awesome.
Open world with instance i heard? Wow nice. I will buy it for sure since is free. 1 mmorpg to pay and one mmorpg for free. Mortal online is the one i will pay, GW is the one for free.
I hope we will have world pvp in GW rather then a BG or arena style like the 1st one and i hope it will be more mmorpg then a Co-RPG like the 1st one either.

no open-world PvP announced yet, unfortunately. There will be "battlegrounds" with casual mass PvP, but that's all that was announced. So you can't kick some charr ass in their hometown for what they did to presearing ;P

I think he is mistaken on this. However, you won't see quests that are like in 90% of the mmos, ie no quest markers above an npc's head. Here is an interview from eurogamer about it.
The quest system, for instance, has been completely overhauled, and will offer something quite different from its competitors. "I think I can safely say that you won't see a single exclamation mark floating above a character's head in Guild Wars 2," claims lead designer Eric Flannum.
"We actually don't have a traditional RPG/MMO quest system," Flannum continues. "Instead what we've got are Events. Think of them as group-orientated activities. This is one of the many things that will encourage the player to explore the world - you can wander through and never quite know what you're going to see. You might come across a fortress that's being attacked by centaurs, or it might be that the centaurs attacked half an hour before you got there and they hold it now. You might start walking along a road you've walked a hundred times and suddenly there's a caravan travelling along that road that you may not have seen, and you can go help that caravan out."

Exactly, no quests in the way people think when you use the term "quest."
No one is going to tell you to go to their farm and kill ten of the wolves scaring the livestock, or whatever. Instead, stuff will be happening in real time in the persistent world. You can participate in this "stuff happening." There will be missions, but we are not clear on what those are. They may be similar to missions in the original which feel like storyline quests in other games, but with more scripting and larger impact on the game world at large. It will be interesting to see what they come up with. The contention over points of semantics is not really necessary, though.

Thanks for the link Cablespider. The event system in combination with the new companion system makes me greatly worried that the game simply cannot be played alone at any time. While playing alone sort of defeats the purpose of the MMO in MMORPG, it's nice to be able to complete small tasks without being reliant on some number of other people being interested in doing the same thing at the same time in the same place. These events all seem raid-like in their nature and I'm very concerned that my event experience will be poor, not just because some players aren't the greatest or others are cheating, but because there weren't enough people at the time (or far too many!). It's also clear from that page that it will be difficult to ask your guild members for help in an event, because if they're not there already, they're not going to make it in time. E.g., the phrasing seems to pretty strongly indicate that they've eliminated map travel, though they never come out and say it. I'm sure events are repeated on regular cycles, but the idea of having to wait several hours to several days for the event to repeat and friends to show up every time I want to do something sounds not fun to me.
In short, I'm very concerned that (1) I won't ever be able to do anything alone, (2) I'll want to participate in an event and it won't be fun because there aren't enough people or too many people, and (3) my ability to play with friends and guildmates will be severely diminished. Hopefully the 'followup' event described in that article will increase or decrease the challenge so that the series of events scales to the people in the area at the time. But I don't think this would alleviate all of my concerns. On the other hand, the feeling of immersion which events will give should be pretty darned cool.

Taken from the GW2 FAQ!
Will Guild Wars 2 be solo-able?
Yes. You will be able to advance your character to the maximum level without ever joining a group if you so desire. Most content will be designed in a solo-friendly way, though often with mechanisms for scaling up in difficulty when more players are involved. This will give players the option to experience the game however they prefer.
At the same time, it is important for an MMO community to join together to overcome challenges. Guild Wars 2 will feature challenges that require players to join forces.

Thanks majinant, that's good news! It still doesn't alleviate my concern about playing with friends and guild members, but I appreciate that they won't forget about the 'loners.' :-)

Hopefully this will be different, we all know the WAR turned into a giant hype machine that barely lived up to its expectation but guild wars 2 seems to be going about its business in a far different way. I mean their whole concept of the release date is "when it's ready". Personally I think that speaks volumes about a company that is willing to hold back a couple of months for dev. sake and isn't just seeing big dollar signs.
