I am a software engineer and I could never use a stand up desk. Left knee was reconstructed in 1984 and needs replacement, right knee and hip are messed up now, arthritis in major joints. If I stand for too long, left leg goes numb.

The problem isn't sitting as opposed to standing. Standing still for hours on end is probably worse than sitting still. The problem is holding one posture for hours without moving around much; the only such posture that is healthy is laying down while sleeping, and even then, only if you don't lay down that much during the day.
It is easier to move around while standing than sitting. I haven't tried it, but I don't think the ergonomics of standing while using a mouse would be very good. Of course, a lot depends on what you're doing at a computer; 8 hours per day of aggressive mouse usage is asking for trouble no matter what your posture is.
There are desks that go up and down so that you can stand part of the day and sit part of the day. That might work pretty well, though I haven't tried it. I'd rather just get up and walk around periodically, which doesn't require any special desk accommodations.
Well I make my own stand up desk and it does really make a difference. Stop having back problems and burn some fats too . My leg feels stronger and better.
