Cinderella man - "I'm being good. I'm being quiet. I'm being have'. James Braddocks son. Love it because it is such a simple line but coming from a small child is so perfect. He has obviously been told to be quiet, be good and to behave but such a good take on a child's perspective.
Shawshank redemption - "I guess I just miss my friend". Andy Dufresne. A simple line that gets to the heart of the story, friendship can overcome any adversity.
A time to kill - "now imagine she was white". Jake Brigance. Again this line just jolts you when you hear it. I cannot think of a better film for displaying the equality of all people and the dis-equality that continues for really pathetic reasons. The entire film builds to this one simple line and boy is it powerful.
Aliens - "Game over man, game over!". Hudson. This line is perfect for the film. When the cocky guy finally realises they are screwed.
I watch many films and I have so many favorites but I am always open to watching new films to add to my list. So with the hopes of expanding my list...
Do you have favorite lines from films and if so what are they?

“You're only supposed to blow the bloody doors off!” - Micheal Caines most famous/repeated liner
this is just a few but im sure i could think of 100s, heck could almost quote every line in airplane... but my number one goes to
"I'm gonna make him an offer he can't refuse." - Godfather, which in all honesty has gone down into normal day of life
but on the airplane, then the best one liners with different endings goes to lloyd bridges

Hawkins: A jester unemployed is nobody’s fool!

Hawkins: I’ve got it! I’ve got it! The pellet with the poison’s in the vessel with the pestle; the chalice from the palace has the brew that is true! Right?
Griselda: Right, but there’s been a change. They broke the chalice from the palace.
Hawkins: They broke the chalice from the palace?!
Griselda: And replaced it with a flagon.
Hawkins: A flagon?
Griselda: With the figure of a dragon.
Hawkins: Flagon with a dragon.
Griselda: Right.
Hawkins: But did you put the pellet with the poison in the vessel with the pestle?
Griselda: No! The pellet with the poison’s in the flagon with the dragon! The vessel with the pestle has the brew that is true!
Hawkins: The pellet with the poison’s in the flagon with the dragon; the vessel with the pestle has the brew that is true.
Griselda: Just remember that.

From "How to Murder your Wife"
Charles Firbank: Good God. Doesn't speak English? And yet, on the other hand, if one will go around marrying persons who pop out of cakes, it's bound to be, well, rather catch as catch can, isn't it, sir?

Favourite quote / one liner, a tough nut. Kevin Smith used to write great lines (mostly for Jay and Randall), same for Tarantino in his early movies (usually for Tim Roth, especially as the bellhop ), Mel Brooks and the ZAZ trio for comical one liners, and of course almost all the sketches and movies from the Pythons...
But, since @turtle mentioned Mononoke Hime, I think I'll drop in a great part from his masterpiece, Naushika:
Look at my hand. I have the same disease as master Jiru, in six months my hand will be stump like a rock. But she said my hand is beautiful... The hand of a hard-working man.
(not in brackets, 'cos never saw it with english dub... but something like that)
Count Rugen: Stop saying that!
Inigo Montoya: HELLO! MY NAME IS INIGO MONTOYA! YOU KILLED MY FATHER! PREPARE TO DIE!
Inigo Montoya: Offer me money.
Count Rugen: Yes!
Inigo Montoya: Power, too, promise me that.
Count Rugen: All that I have and more. Please…
Inigo Montoya: Offer me anything I ask for.
Count Rugen: Anything you want…
Inigo Montoya: I want my father back, you son of a bitch!
edit: should say it's one of my favorite films and I own a copy. I make sure i see it once per year along with some of my other favorites such as the "How to Murder your Wife", "Bell Book and Candle," "Rear Window" and the 1958 George Pal "War of the Worlds".

Captain Oveur: I can't tell.
Rumack: You can tell me. I'm a doctor.
Captain Oveur: No. I mean I'm just not sure.
Rumack: Well, can't you take a guess?
Captain Oveur: Well, not for another two hours.
Rumack: You can't take a guess for another two hours?
