I will argue until my last breath that the below should be included in any greatest games lists:
...Tons of Games... Overwatch ...Tons of Games...
I realize it's a bit shallow but I saw a gorilla in glasses in all the Overwatch promo material and that is the main reason I never tried the game. Just seemed targeted directly at kids to me. Mind you there are things just as ridiculous in games that I play and love but they weren't the first impression I ever got of the game.
There's a free week-end coming up on November 18th for Overwatch so you should check it out if you like this kind of FPS (like Team Fortress), it's a fun game. It's for a general audience and Blizzard put a lot of effort into fleshing out all their different characters with nice backstories, the animated shorts they've made so far for some of the characters are really great.
I wanted to add Dishonored yesterday but after dropping in to Twitch i see Dishonored 2 is out ..:P Imo a great game now series,way under rated,i couldn't stop playing unti l ifinsished it then played theaddons and went back and played again and again.Now i see number 2 is out or near out and i can't wait to play it.

Not a lot of people realize that Wurm Online is Minecrafts daddy, it was Notch's baby. IMO its one of the true sandbox mmo's out there, forget SWG or BDO or any of these so called sandbox mmo claiming to be sandbox.
For me this question brings back memories from my childhood. So here is my list:
For PC: It has to be MU online, MU legend, Ragnarok, Counterstrike, Dota and Dota 2, StarCraft and warcraft lastly WoW.
For Mobile: MU Origin, Clash of Clans, Pokemon Go and Summoner's War.
For Console: Mario Bros 1/2/3, Megaman, Sonic, Tekken, Mortal Combat, King of Fighters, ninja gaiden, Onimusha, God of war, DMC, GTA, Final Fantasy, NBA, and lastly Pac-Man
JRPG: FFVII, it may not be the best JRPG, or even the best of the Final Fantasies, but there's no denying it's impact on the genre and gaming in general. Bravely Default gets a mention here as well.
Western RPG: Dragon Age Origins, or Morrowind - I could go either way. Both of those games made a big impression on me.
RTS: Starcraft is probably the definitive one, I would also mention Homeworld, as it left a large impression on me.
Racing: I have to agree with Mario Kart on this one. Pretty much any of the series.
Isometric - Diablo is the defining game here, but I'd also like to mention Bastion/Transistor here as well.
Turn-based Strategy - Civilization... any of the series really. I probably have the most hours on 3. Alpha Centauri definitely deserves to be mentioned here, as well as X-Com
FPS: Doom really kicked it all off, Quake took Doom and cranked it up. Then it went multiplayer, and, at least for me, that becomes a tossup between Tribes and Unreal Tournament. I'm sure a younger gamer may say CoD or BF or Halo or Overwatch, but for me, those older games were all definitive.
MMO: WoW (another Blizzard title again) defines the genre today, for better or worse. EQ1 gets a mention here too, because that was what WoW more or less was based on. I'd say SWG and CoH were the real must-play MMOs, but alas...
PVP/Competitive - Either Street Fighter 2 or Eve Online. Both are totally different games, and compete in totally different ways. But if you are just looking at matching yourself up against other people, these are the 2 titles that made the biggest impression on me.
SimCity 2000. Not really sure what genre this would fall under, it's not really a sim, it's not really turn based. It was just awesome fun. Factorio scratches that itch for me now, and is super fun in much the same way.
Star Wars: Tie Fighter. An oldie, but still very good. My favorite space/flight combat game by far. Privateer needs a mention here too.
Zelda and Mario also need to be on a Must-Play list. Doesn't really matter which title of their respective series you play, they all are significant cultural icons and every gamer should have some exposure to them.

wow.. you are actually asking which games to play o.o i have about 4 tb of games i want to play, but my life span is not long enough to play it all even once, now , many here will subjectively tell you this or that what to play ,some are right ,some are not
from what i can tell you ,most compelling and interesting one of a time experience games are RPG's old and new ones, things like baldurs gate II, deus ex, divine divinity, warcraft III +Ft ,hitman, gothic 2 and 3, star wars jedi academy, fear, half-life 2, star wars republic commando, painkiller , Kotor 1 and 2, thief 3, spell force , titan quest, Nfs , stalker , elder scrolls morrowind + oblivion + skyrim ,witcher series, bioshock, old call of duty and medal of honor games, fallout series, mass effect series ,risen, disciples 3, starcraft 2, sid meier's simulation games any, fable , freelancer, dawn of war soulstorm and few others, riddick ..
if i didnt play all of these, even several times, and i knew how good they are, id jump off a high cliff, in water

Freelancer was one of the best games ever for it's time. It's showing it's age now and the multiplayer servers are mostly dead, but it was an amazing game to experience during it's height.

I'll not list games that are my all time favorites or classics - or anything on that regards.... but I'll (try) to list games I think will blow your mind and change the way you see the world or you see gaming . In my humble opinion of games for over 20 years or so these are games which you can not skip and definitely should at least watch a full (silent if possible) playtrough if the genre is not for you:
- Journey - Child of Light - Undertale - Brothers: A Tale of Two Sons - The Walking Dead (Season One) - A Telltale game - Inside - Machinarium - Botanicula - Braid - I Have no Mouth, and I Must Scream - Life is Strange (yes, even with the plot holes) - Gone Home - Her Story - Devil Daggers - Myst - Riven - Ori and the Blind Forest - The Talos Principle - SUPERHOT - Valley - Crypt of the Necrodancer - The Binding of Isaac - Owlboy - Tormentum - Dark Sorrow - Claire - That Dragon, Cancer
All titles above changed my life a little (some a lot) - just like when you finish a good book or a good movie and you feel kinda empty - due it's core gameplay design, the story telling or aesthetics. But, if you are asking for a single title, and not a list, the answer is simple, short, and predictable: Witcher 3.

@Nyctelios Let me know once you've played them. Both can be found dirt cheap and I wouldn't be surprised if during a Steam sale you could get both for less then a tenner total. To the Moon pulls at your heart strings but Lisa, Lisa does things on another level. Don't let its graphics detract you, it is beautiful in its own right.
I'll be replaying I Have No Mouth soon, looking forward to it already.

Arma 3 w. Sa-Matra Wasteland mission file (has its own launcher software, that handles mod files, via Steam).
Basics: Log onto server, download mission file of about 3MB, pick one of the three factions with up to 80 players or so on the server, join the game and spawn in a town, gear up with stuff found in vehicles or maybe crates, find food/water, go alone or group up with faction membes, go fight ai and players, everywhere, no place is safe! Move around on a HUGE map, on foot, in vehicles, in helicopters, tanks.. Looting stuff and gearing up feels great and empowering, for a well paced game, because you don't simply end up driving around in a tank, like with other variants of Wasteland.
Fire Emblem: Path of Radiance for sure. It's the best Fire Emblem, if not the best Nintendo game of all time. The difficulty is perfectly balanced on Hard. It takes the player through a healthy range of scenarios. All characters are viable on all difficulties. The characters are almost universally likeable. The dialogue and support conversations are the most memorable in the series. The shapeshifter races have actual depth beyond the typical "it's a little girl that turns into a dragon." The Black Knight is the most interesting villain in the series. The soundtrack is stellar. It's just an all-around phenomenal game. Fire Emblem belongs on the consoles, and this title is proof.
This game had lot of stick but in my opinion is the best Fantasy PvP game. Unfortunately it is only fun when there are lots o people playing it and actually since it has been shut by Aventurine, at the moment you can't play it at all. But 2 separate companies are attempting to revive it so if you missed the first round, you should definetly try Darkfall when it re-launches.

Whilst there are many games from my childhood that I'd like to recommend, in reality old games just don't have the same impact to modern gamers. Very few seem to be able to stand up to modern expectations, even if you overlook the graphics. You also can't appreciate how groundbreaking some things were as a modern gamer if you haven't lived through a period of time without such things.
That said, there are a few games I would put on the list, mostly already listed here:
1) Final Fantasy 7 The gameplay is just as good now as it was back then, the story is great and the world and it's inhabitants are awesome. Usually takes me an hour before I start to not notice the bad graphics, but from then on it is just as good as it always was.
2) Age of Empires 2 Still one of my favourite RTSs of all time. I just used to play endless amounts of regicide skirmishes (kill the king) and would just build epic bases. Combat was a bit limiting compared to modern RTS's but it was still a super game. Plenty of upgrades, loads of units (far more than the C'n'C series), some great sound effects and just a thoroughly enjoyable game.
3) Medieval: Total War You can pretty much pick any total war for this list, but I think Medieval shows off the game to it's best. Exceedingly tactical game with so many options, Medieval had possibly the greatest selection of units of all the total war games and the least amount of limitations. You should play the total war series in order to experience the massive RTS battles. 1000v1000 battles (think that was limit in medieval, obv a lot higher in latest release) are just great. Getting your squads into the right formations and layouts is crucial. Total War: Warhammer is probably the best for the actual battles, what with having heroes, magic, mythic beasts etc, but it lacks the tactical map stuff of earlier games.
4) MX Superfly This is a motocross game for original xbox. Possibly the best motocross game I've ever played. Half of it was proper racing, half of it was freestyle tricks, all of it was fun. I've sunk 100s of hours into this game over the years. It has the usual stadium races, outdoor races and the like, as well as stadium freestyle, race freestyle as well as open maps to freestyle in. There were some great easter eggs too, for example, if you followed a long and complex series of hidden jumps on one map, you could enter roswell and have fun in a halfpipe beneath a ufo.
5) Skate 3 One for the 360 / PS3 crowd. Best skateboarding game ever made. The trick system is just awesome and everything you do looks great. 3 was a little easier than 1 or 2, but had more tricks and more fun stuff to do. "Hall of Meat" was great - basically, stack it and break as many bones as possible. Also had a really fun park editor, though being EA they did put half the stuff in a DLC. Really wish they'd bring the game to PC so we could mod it!
Honorable mention to: Sega Worldwide Soccer 97! Only football game I've ever enjoyed, the controls actually worked as advertised! Best bit about it was that the commentators were really badly programmed so were constantly saying stuff that had no bearing on what was happening. Constantly kept me laughing! This was on the Sega Saturn though, so might be a bit hard to try it out these days.....

Super Smash Bros (any, but prob Melee is best). This game elevated couch gaming with buddies to a whole new type of experience.
