My best quest ever and probably the reason I have not forgotten that after many years was in Runescape and it was a quest with a bunch of doors that you had to know the combinations of locks to do the quest which they were absolutely impossible to guess. The part that made that the best quest for me was I figured out the combination and I was sitting in front of the entrance door for a while charging newbies a hefty amount to teach them how to do the quest. It was so much fun
For any quest, I'd have to say Skyrim's Dragonslayer quest or the Defense of Whiterun. If you do defense of whiterun on hardest difficulty, it's like an all out war with like 100 guys charging the gates
Tbh in general Skyrim's quests related to dragon fights or main plot are more fun than in most other RPG games.
As in topic never really thought about it but if I had to choose something then it probably would be any having nazguls in LotRO.
So I like many others love Chains of Promathia from FFXI, great story, great missions, just all around amazing story arche.
I will get a bit more specific than that. 6-4 one to be feared. I will never forget that quest/mission whatever you want to call it.
It was one of the hardest mission I've had to deal with in an MMO and before nerfing there was no trick or cheat to make it easy. It was all about strategy and team work, none of this zerg zerg zerg crap that took over the game later on. Not to mention it was just epic battling in the middle of all those airships.
This was based on sense of accomplishment. There is this shawl in EQ that gives +8 to alot of stats. Basically its for the shoulders and good for pretty much any character. Quest is a pain in the but, requires you to get alot of skills up like fishing and sewing. Real pain, but sense of accomplishment was worth it. I completed that quest for my enchanter who was 65th when I left the game. Haven't been back since then, guess they raised max level up some and this item isn't as uber.
In Tabula Rasa some toughguy soldier sent me out to a tank in the middle of a battlefield to recover a porno mag he had left there. In addition to being funny, it was incredibly immersive and perfectly fit into the type of world that you were part of. Still stands out as one of the best quests I've ever done.
Another example is in being sent to reforge The Blade That Was Broken. Perfectly representative of that game's amazing ability to put the player "on the side" of the fellowship but into meaningful and fascinating situations storywise.

This was my favorite, followed by the wizard epic, click stick and many others from EQ1.
